Moonwalk Biosciences Raises $70M for Obesity RNAi Pipeline

Moonwalk Biosciences has closed an oversubscribed $70M Series B to move an adipose-targeted RNA interference program toward the clinic. Alpha Wave Global and YK Bioventures co-led the September 8, 2026 financing, with Eli Lilly and Company, Gaorong Ventures, ARCH Venture Partners, Khosla Ventures, and Future Ventures participating.

The capital will support IND-enabling work for MW101, an experimental siRNA medicine for obesity, and a first-in-human study targeted for late 2027. It will also fund additional tissue-targeted siRNA programs and Moonwalk's discovery platform, which combines human genetics, epigenomics, multi-omics analysis, and adipose-selective delivery chemistry.

The round matters because Moonwalk is narrowing a broad platform thesis into a specific clinical bet. The company started by mapping and engineering the epigenome, used that work to identify adipose targets, and then chose RNAi as the modality it believes can move those targets toward patients faster.

What Moonwalk Biosciences' $70M Series B Funds

Moonwalk's financing announcement describes the Series B as closed and oversubscribed. Alpha Wave and YK Bioventures co-led, while Lilly and Gaorong joined returning investors ARCH, Khosla, and Future. The company did not disclose its valuation, ownership terms, individual check sizes, or cash runway.

Moonwalk publicly launched in January 2024 with $57M across Seed and Series A financing. The new round brings disclosed capital across the 2 announcements to $127M. That figure is not a single financing and does not include any undisclosed capital.

The immediate work is preclinical and regulatory. Moonwalk plans to complete ongoing IND-enabling studies for MW101, prepare a first-in-human study for late 2027, and advance additional siRNA candidates aimed at other mechanisms in adipose tissue. The company also plans to continue developing the discovery system that produced the targets behind the pipeline.

Why Moonwalk Chose RNAi for Its First Clinical Push

Moonwalk launched around EpiRead and EpiWrite, technologies designed to map epigenetic cell states and perturb gene regulation without changing the underlying DNA sequence. Its current science platform layers human genetic associations over epigenetic differences in white, brown, and precursor fat cells, then tests proposed targets in disease-relevant models.

The output of that work did not obligate Moonwalk to use epigenetic editing as the first therapeutic modality. CEO Alex Aravanis told BioPharma Dive that RNAi offered a faster route to the clinic for the targets the company found. Moonwalk exclusively licensed adipose-targeting siRNA chemistry from Suzhou Siran Biotechnology and built its first pipeline around that delivery system.

That is an important platform-company decision. Discovery technologies are often marketed by the number of targets or diseases they might reach, but drug development eventually requires a company to choose a target, delivery route, candidate, and clinical sequence. Moonwalk's first large test is whether its discovery work can make that narrowing more informed.

How MW101 Approaches Obesity Differently

MW101 is designed to deliver a small interfering RNA directly to adipose tissue and modulate a novel non-incretin pathway. Rather than suppressing appetite, Moonwalk says the program is intended to alter biology inside fat cells. The wider pipeline explores mechanisms connected to lipolysis, adipogenesis, thermogenesis, energy homeostasis, and adipose endocrine function.

Moonwalk reports that its siRNA programs reduced body weight and fat mass in preclinical models while preserving lean mass and without reducing food intake. It also reports durable adipose target engagement after one dose in non-human primates, evidence the company says could support quarterly or twice-yearly dosing. These are company-reported preclinical findings, not human safety or efficacy results, and MW101 has no disclosed clinical-trial identifier.

The distinction matters because incretin medicines have transformed obesity care while leaving room for different mechanisms, combinations, dosing schedules, and body-composition profiles. A 2025 systematic review indexed by PubMed found that GLP-1-based medicines reduce fat mass and can also reduce absolute lean mass, although results vary by agent and study. Moonwalk's claim that it can preserve lean tissue in people will have to be established in clinical development.

Why Eli Lilly's Participation Draws Attention

Lilly's investment puts an incumbent obesity-drug developer inside the syndicate for a non-incretin program. That makes the round strategically interesting, especially as drugmakers look beyond appetite suppression toward complementary mechanisms and combination strategies. The announcement, however, describes Lilly only as an investor and discloses no license, option, development partnership, or acquisition right.

The rest of the syndicate also reflects continuity. Alpha Wave and YK supported Moonwalk's 2024 financing and returned as co-leads, while ARCH, Khosla, and Future invested again. Their follow-on capital is a vote to keep funding the scientific path, but it does not settle the program's clinical questions.

The Team Carrying MW101 Toward the Clinic

Moonwalk was co-founded by CEO Alex Aravanis, President and CTO Arash Jamshidi, Chief Platform Officer Justin Valley, and scientific co-founder Feng Zhang. The current leadership team also includes Chief Business Officer Elaine Cheung and VP of Biology and Translational Science John Liles.

The Series B announcement adds Stephen Djedjos as SVP and head of clinical development. Djedjos most recently led clinical development at Kailera Therapeutics and previously held roles at Mineralys Therapeutics, Sana Biotechnology, Gilead Sciences, and Amgen. His appointment puts an experienced clinical operator beside a program whose next milestones are increasingly regulatory, translational, and operational rather than purely computational or discovery-driven.

The Clinical Handoff This Capital Is Buying

Moonwalk has assembled a differentiated thesis: adipose tissue can be targeted directly, epigenetic and genetic evidence can improve target selection, and siRNA may offer durable activity without depending on appetite suppression. Each part remains a hypothesis until MW101 produces a regulatory package and human data.

The $70M Series B gives the company resources to make that handoff. Over the next stage, Moonwalk must turn its platform evidence, licensed delivery chemistry, preclinical pharmacology, and new clinical leadership into a candidate ready to enter people. The company has selected the tissue and modality; late 2027 is where that selection is expected to meet its first human test.
