SereNeuro Is Turning Pain-Sensing Neurons Into Therapy

SereNeuro Therapeutics is a Baltimore biotechnology company developing non-opioid therapies for chronic pain. Its lead program, SN101, uses pain-sensing neurons derived from induced pluripotent stem cells to address chronic osteoarthritis joint pain. A second program, SN102, uses gene therapy to target neuropathic pain.

Dan Saragnese, CEO and co-founder, leads SereNeuro with Chief Scientific Officer Laura Richman, Chief Technology Officer Oliver Kraemer, and Senior Principal Scientist Tea Soon Park. Scientific co-founders Xinzhong Dong, Gabsang Lee, and Zhuolun "Poppy" Wang connect the company to the Johns Hopkins research that produced its core biology.

SereNeuro matters because its lead asset reverses the usual logic of pain medicine. Instead of only blocking pain-sensing neurons, SN101 adds mature nociceptors to an arthritic joint and asks those cells to absorb inflammatory pain factors. That mechanism remains preclinical, but it gives SereNeuro a distinct place in the expanding search for non-opioid pain therapies.

About SereNeuro Therapeutics

SereNeuro traces its origin to a research model built to study human peripheral pain. Xinzhong Dong, Gabsang Lee, and Wang created human nociceptors from induced pluripotent stem cells so researchers could investigate pain targets in a more relevant system. The cells were a tool, not a treatment plan.

The direction changed when the team placed the cells into a joint model and saw an unexpected therapeutic effect. The observation created the foundation for SN101 and for a company that LinkedIn lists as founded in 2024. Saragnese joined the scientific founders to build a development organization around that result.

That origin is more than a memorable company story. It explains SereNeuro's operating challenge. A research model can be valuable because it behaves like human biology. A therapy has to do that repeatedly, at a defined dose, with a manufacturing process and safety profile a regulator can evaluate.

SN101 Uses Living Cells as a Local Pain System

SN101 is an off-the-shelf therapy made from purified human nociceptors. SereNeuro says the cells express the channel identity associated with native dorsal root ganglion neurons, including Nav1.7, Nav1.8, and Nav1.9. The candidate is delivered into an affected joint, where the cells are intended to interact with the inflammatory environment without relaying those pain signals to the brain.

The company describes a dual mechanism. SN101 cells may sequester inflammatory pain factors and release regenerative factors associated with cartilage preservation. SereNeuro presented preclinical findings through ISSCR showing pain relief and reduced cartilage degeneration in models. Independent reporting says the company has also conducted primate work and studied osteoarthritis joint-fluid samples.

Those results support further development, not a conclusion about patients. SN101 has not produced human clinical safety or efficacy data. The company is conducting IND-enabling work, where cell identity, purity, maturity, dose, persistence, delivery, and manufacturing consistency become inseparable from the therapeutic claim.

SN102 Extends the Platform Beyond Osteoarthritis

SereNeuro's second program approaches pain through a different modality. SN102 is an in vivo gene therapy in lead optimization for trigeminal neuralgia and other neuropathic pain. The program is designed to reduce hyperactive nociceptor signaling while preserving ordinary sensation and motor function.

The National Institute of Dental and Craniofacial Research awarded SereNeuro a $318,000 Phase I Small Business Innovation Research grant under the NIH Helping to End Addiction Long-term Initiative. That award supports SN102 and is separate from the company's pre-seed financing.

Together, the programs make SereNeuro more than a single unusual cell-therapy experiment. SN101 tries to change the local environment around chronic joint pain by adding new cells. SN102 tries to adjust the electrical behavior of existing sensory neurons. Both are preclinical, and each carries different delivery, manufacturing, safety, and regulatory questions.

Leadership Is Moving the Science Toward Development

SereNeuro's team combines pain neuroscience, cell engineering, gene therapy, and biotechnology operations. Dong and Lee bring decades of Johns Hopkins research in sensory biology and stem-cell-derived neural systems. Wang helped build the research platform that became the company's starting point.

Saragnese has experience moving therapeutic programs from preclinical work into clinical development at Johns Hopkins and Stanford spinouts. Richman brings biotechnology leadership and development experience, including work in regenerative medicine and bioprocessing. Kraemer leads technology, while Park leads cell-therapy science at the point where experimental biology has to become a controlled product.

The company recently recruited for a Chief Scientific Officer and now lists Richman in the role. That completed hire is a more useful signal than generic hiring language. SereNeuro is building the technical leadership needed to prepare a cell therapy for regulatory review. No current public list of open roles was verified, so the evidence supports team expansion, not an active hiring claim.

Maryland Is Part of SereNeuro's Development Strategy

SereNeuro operates from East Baltimore near Johns Hopkins research, clinical, and commercialization infrastructure. The company has received support from Maryland biotechnology programs and participated in the state's BIO 2026 delegation. Its pre-seed investors also used Maryland's Biotechnology Investment Incentive Tax Credit, according to the company.

That geography matters for an early cell-therapy company. SereNeuro needs scientific collaborators, translational facilities, regulatory experience, specialized talent, and investors willing to finance development before clinical proof exists. Maryland gives the company proximity to those resources while keeping the work connected to the laboratory that produced it.

SereNeuro announced an oversubscribed pre-seed financing on October 5, 2026. The company did not disclose the amount, valuation, or investor names. It said the capital and non-dilutive funding would support manufacturing and preclinical work for SN101.

What SereNeuro Signals for Non-Opioid Pain Development

Chronic pain is a systems problem. Inflammation, damaged tissue, sensitized peripheral nerves, and changes in central processing can reinforce one another. A single receptor blocker may affect one part of that loop. SereNeuro's thesis is that living cells can interact with several local signals at once and potentially support both pain relief and joint preservation.

That breadth is attractive, but it increases the proof required. A durable cell therapy must behave predictably after injection, avoid unwanted sensation or growth, remain consistent across manufactured batches, and show that an effect in animal models can translate into meaningful patient benefit.

SereNeuro is still before that clinical test. What it has today is a differentiated biological idea, a team grounded in the relevant science, two distinct preclinical programs, and new capital to move the lead candidate through harder development gates.

The larger signal is that non-opioid pain innovation is moving beyond small molecules and device-based symptom control. SereNeuro is asking whether engineered human neurons can become part of the treatment itself. The company will be judged by the evidence it produces next, but the question is serious enough to follow.
